Role of cyclic nucleotides in the proliferation of committed granulocyte-macrophage progenitor cells.

چکیده
The proliferative response of murine hemopoietic pro genitor cells to the stimulatory actions of colony-stimulating factor can be antagonized by agents that elevate cyclic adenosine 3':5'-monophosphate and can be potentiated by agents associated with increases in cyclic guanosine 3':5'monophosphate. The granulocyte-macrophage progenitor cell is profoundly inhibited by the E-series prostaglandins, whereas its proliferative ability can be augmented by low concentrations of prostaglandin F._,«. The existence of differ ent receptors for prostaglandins E and F, determined with the prostaglandin antagonist, SC-19220, correlates with the differential effects of the two prostaglandin subclasses on colony-stimulating factor action. Tumor necrosis factor (TNF)-alpha directly inhibits human erythropoiesis in vitro: role of p55 and p75 TNF receptors.
Two tumor necrosis factor receptors (TNFRs) with molecular weights of 55 kD (TNFR-p55) and 75 kD (TNFR-p75) have recently been identified and cloned. In previous studies, TNFR-p55 has been shown to exclusively mediate bidirectional effects of TNF-alpha on committed bone marrow granulocyte-macrophage progenitor cells, whereas both TNFR-p55 and TNFR-p75 can mediate inhibition of primitive progeni...
متن کاملHematopoietic progenitors in cyclic neutropenia: effect of granulocyte colony-stimulating factor in vivo.
The number and growth factor requirements of committed progenitor cells (colony-forming units-granulocyte/macrophage and burst-forming units-erythroid) in three patients with cyclic neutropenia (two congenital, one acquired) were studied before and during therapy with recombinant human granulocyte colony-stimulating factor (G-CSF; 3 to 10 micrograms/kg/d). When the patients with congenital dise...
متن کاملCYCLIC NUCLEOTIDES CONTROL DIFFERENTIATION OF HUMAN MONOCYTES INTO EITHER HIGHLY ACCESSORY CELLS OR MACROPHAGES
Human peripheral blood monocytes have been found to undergo a transitory state of high accessory activity before they fully become macrophages. Time kinetics were done to follow this accessory potential. Studying the regulation of accessory activity, we have found that monocyte derived accessory cells (m-AC) pass through two phases of development, both of which are adversely controlled by ...
متن کاملOverexpression of Recombinant Human Granulocyte Colony-Stimulating Factor in E. coli
Bakground: Granulocyte colony-stimulating factor (G-CSF) is a cytokine that stimulates hematopoiesis and induces proliferation and differentiation of granulocyte progenitor cells as well as production of bone marrow neutrophilic granulocyte colonies. Nowadays, human recombinant G-CSF(hr G-CSF)is used for the treatment of chemotherapy- and radiotherapy-induced neutropenia, and also in patients ...
متن کاملAbnormal Responses of Myeloid Progenitor Cells to Granulocyte - Macrophage Colony - stimulating Factor in Human Cyclic
Granulocyte-macrophage progenitors (CFU-GM) from four patients with childhood onset cyclic neutropenia demonstrated abnormal in vitro proliferative responses to purified, recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) when examined in detailed dose-response studies. Marrow aspirate specimens were obtained for these studies from cyclic neutropenia patients (both du...
